• Custom DNA Sequencing services by Synergene Biotech combine leading technologies of this field. We perform AB 3100-based fluorescent sequencing and apply Big DyeTM v.3 terminator chemistry. We offer two kinds of services:

• Please note: For both runs, no editing, no repeat of a run, invoicing irrespective of the results. Prices given do not include 7.6% VAT and shipping charges. Placing an order implies that you agree with our terms and conditions provided here. For orders of a certain size, special terms and conditions could apply.

• Please send the Template DNA in 200 ul PCR tubes or 8-well stripes, or 96-well microplates. Due to automatic processing, whenever possible use tubes or stripes from AB-Gene available for example at http://www.biolabo.ch/frameset_contacts.htm
• Dissolve the DNA in 15 ul ddWater (not TE buffer). Use the following reference concentrations:

  Plasmids < 5 kb	60 ng / ul
  Plasmids > 5 kb	150 ng / ul

  
  
• If needed, add 1 ul Primer solution (10 pmol / ul). You can use your own primers or choose a standard primer to be added by Synergene Biotech (service is free of charge). See our primer list for details.
• Fill out the submission Excel File (download here).
  Attach this file to an email message and send it to sequencing@synergene-biotech.com . Please remember to save the file before attaching it, otherwise we will not be able to read it.
• Send us your samples by A-post (inside Switzerland) or any other fast shipping service when ordering from another country. For your convenience we can provide addressed padded bags and tubes upon request.

• The following tips are drawn from practical experience. They can be helpful whenever commercial kits from suppliers such as Promega, Qiagen, etc. are used to purify DNA templates:
  
• Make sure you do not overload the columns with bacteria when extracting plasmid DNA since in such cases, the process of purification tends to be less efficient.
  
• Both PCR and plasmid purification work best if you do not try to recover the eluate down to the very last drop. Commercial kits yield DNA of excellent quality but overelution can put unwanted dirt back to your sample, which leads to poor sequencing results. Since commercial protocols have been extensively tested, it is recommended to follow them exactly.
  
• Please make sure that no EDTA residues are left inside your sample. Some commercial kits use elution buffers containing EDTA, such as TE buffers. EDTA complexes magnesium ions, which reduces the effectiveness of the sequencing polymerase. The results of your sequencing can therefore be severely compromised. Please contact us if your protocols include steps using EDTA based buffers.
  

• Depending on your platform you can view the resulting trace files with the following software:
  
  Windows: Chromas (download at http://www.technelysium.com.au/chromas.html)
  
  Mac: Edit View (download at http://www.appliedbiosystems.com/support/software/dnaseq/installs.cfm)
  
  
• Notice that as a Mac user you will need to convert the trace files into a suitable format for Edit View. The conversion utility can be downloaded at: http://www.appliedbiosystems.com/support/software/3100/utilities.cfm.
  
  To convert the trace files to Mac format, please proceed as follows:
  
  1. Create a new folder in any location you find appropriate.
  2. Copy the trace files into the new folder.
  3. Double click on the icon representing the conversion tool 'Win to Mac'.
  4. Navigate to the location from where you can see your new folder.
  5. Click on it to select it. Make sure you click only once (do not enter the folder).
  Notice: At this point you will not be able to see the file names inside the folder.
  6. Click on the 'choose' button on the rightmost side of the window.
  7. After a few seconds, you should be able to open the files with Edit View.
